Intended purpose

The assay attomol® Factor XIII V34L Quicktype is used for the supportive diagnosis of thrombophilia. This test can be used to determine the transversion 102G>T in the human factor XIII gene (rs5985). It is a manual, qualitative assay using an allel-specific PCR with integrated probe hybridisation. The evaluation takes place by agarose gel electrophoresis, which can be carried out manually or automatically. The sample material has to be genomic DNA prepared from EDTA or citrate blood.

Field of application

A  deficiency  in  the  coagulation  factor  XIII  is  an  autosomal  recessive  disease  which  is  characterised  by  a disturbed healing process, tendency to spontaneous bleeding and an increased risk of miscarriages in the case of  affected  women  [Mikkola et al., 1994, Blood, 84:517-525].  Factor  XIII  activated  by  thrombin  ensures  the  cross-linking  of  fibrin  which  is  very important for the stability of formed blood clots [Undas et al., 2005, J Thromb Haemost, 3:1-7].One polymorphism often named in connection with factor XIII is the base exchange of G to T in codon 34 which causes an amino acid substitution of valine to leucine (Val34Leu) on the protein level. This mutation is only 3 amino acids away from the thrombin activation point and thus it leads to an easier activation of factor XIII. This fact could play a role in the development of thrombotic diseases [Kohler and Grant, 1999, Q J Med, 92:67-72; Wiwanitkit, 2005, Haema, 8:318-320]. The mutant allele is reported with a prevalence of about 20-45% for the Caucasian population. The prevalence of mutation varies among different ethnic groups [Wiwanitkit, 2005, Haema, 8:318-320].
