Intended purpose

The assay attomol® Fibrinogen beta Quicktype is used for the supportive diagnosis of thrombophilia. This test can be used to determine the transition -455G>A in the human fibrinogen beta gene (FGB gene, rs1800790). It is a manual, qualitative assay using an allel-specific PCR with integrated probe hybridisation. The evaluation takes place by agarose gel electrophoresis, which can be carried out manually or automatically. The sample material has to be genomic DNA prepared from EDTA or citrate blood.

Field of application

Fibrinogen is a coagulation factor consisting of three polypeptide chains (alpha, beta, gamma), which is converted to fibrin by thrombin. Fibrin is an essential component for the formation of a coagulation thrombus [Hoppe, B., 2014, Thromb Haemost., 112, 649-658]. Abnormalities in this system can lead to an increased risk of thrombosis [de Willige et al., 2005, Blood, 106, 4179-83]. In the review by Iacoviello et al. it is reported on the basis of various publications that transversion -455G>A in
the promoter region of the fibrinogen beta gene is associated with an increased fibrinogen concentration [Iacoviello et al., 2001, Hypertension, 38, 1199-1203].
Also in the editorial of Papageorgiou et al. the mutation -455G>A is described as the most important genetic polymorphism associated with increased fibrinogen concentration [Papageorgiou et al., 2010, Hellenic J Cardiol, 51: 1-9]. Patients with a fibrinogen concentration greater than 5 g/L were observed to have a 4-fold increase in the risk of thrombo-embolism [Koster T. et al., 1994, Thromb Haemost., 71, 719-722].
