Intended purpose
The assay attomol® Factor II 19911A>G Quicktype is used for the supportive diagnosis of thrombophilia. This test can be used to determine the transition 19911A>G (rs3136516) in the human factor II gene (prothrombin mutation). It is a manual, qualitative assay using an allel-specific PCR with integrated probe hybridisation. The evaluation takes place by agarose gel electrophoresis, which can be carried out manually or automatically. The sample material has to be genomic DNA prepared from EDTA or citrate blood.
Field of application
In the second-last step of the blood coagulation cascade, prothrombin (factor II) is converted into the serine protease thrombin. This converts fibrinogen molecules by splitting the fibrinopeptides A and B into fibrin monomeres, which spontaneously aggregate to fibrin polymers. In addition to the polymorphism 20210G>A in prothrombin, 19911A>G was identified as a second polymorphism in the same gene, which is also associated with an increase in prothrombin concentration in blood plasma and thus with an increased risk of venous thromboembolism [Chinthammitr et al., J Thromb Haemost 2006, 4:2587-92]. The mutated allele of polymorphism 19911A>G occurs in the European population with a prevalence of approximately 47 % [U.S National Library of Medicine, SNP-Database, 2019-09-19]. Furthermore, it was found that the 19911A allele is strictly coupled to the 20210A allele [Zivelin et al., Blood 1998, 92:1119-24]. Especially in combination with other polymorphisms and risk factors a determination of the polymorphism' 19911A>G should be considered [Martinelli et al., J Thromb Haemost 2006, 4:2582-6].
