Assay for the detection of the deletion variants GST M1 and GST T1 of the glutathione S-transferase genes.

Glutathion S transferases (GST) belong to the enzymes of phase II turn over. They conjugate electrophile foreign substances or metabolites of them with glutathione. This reaction acts in almost every case as a detoxification process but can also lead to a metabolic activation [Schulz et al., 2002, Umweltmed Forsch Prax, 7(4):232-246]. For the majority of GST, sequence variations (polymorphisms) are known encompassing the variants GST M1, GST T1 and GST P1. The polymorphisms GST M1 and GST T1 are gene deletions that cause the loss of enzyme activity in homozygous patients. The occurrence of these gene deletions is called null genotype. The null genotypes of GST M1 and GST T1 are discussed in the context of certain tumours e.g. bladder cancer [Empfehlung des RKI, Mitteilung der Kommission „Methoden und Qualitätssicherung in der Umweltmedizin“, 2006, Umweltmed Forsch Prax 11(1):55-64]. The significance of GST null genotypes with respect to environmental medical indications and the metabolism of some drugs (chemotherapeutics) is still discussed controversially. The null genotype of GST M1 and GST T1 occurs with a frequency of 50 % and 20 %, respectively in Europe [Schulz et al., 2002, Umweltmed Forsch Prax, 7 (4):232-246; Empfehlung des RKI, Mitteilung der Kommission „Methoden und Qualitätssicherung in der Umweltmedizin“, 2006, Umweltmed Forsch Prax 11(1):55-64].
